What if a Trulicity® (dulaglutide) dose is missed?
If a dose of dulaglutide is missed and there are more than 3 days (72 hours) until the next scheduled dose, it should be administered as soon as possible.
Instructions for a missed dose of dulaglutide
Dulaglutide is a human glucagon-like peptide-1 receptor agonist for once-weekly administration.1
If patients miss a dulaglutide dose, their next dose depends on how much time there is before the patient's next regularly scheduled injection day (Instructions for a Missed Dose of Dulaglutide).1

The day of weekly administration can be changed, as long as the last dulaglutide dose was administered longer than 3 days (72 hours) ago.1
Pharmacokinetics of dulaglutide when a dose is missed
Dulaglutide improves glycemic control after the first dose and maintains the control throughout the once-weekly dosing interval.1
It reaches a steady-state plasma concentration between 2 and 4 weeks following once-weekly administration.1
Once steady state is achieved, missing 2 or more consecutive doses may result in subtherapeutic dulaglutide concentrations (Effect of Missed 1.5 mg Once-Weekly Doses on Dulaglutide Pharmacokinetics at Steady State).1
Figure 1 description: Between the 2nd and 4th dose, dulaglutide reaches its steady-state concentration. When patients missed one dose and took it 3 days before the next regular dose, the dulaglutide concentration was higher in the week of the regular dose, but returned to steady-state when they continued dulaglutide once-weekly. One completely missed dose resulted in lower dulaglutide concentrations, which returned to steady-state in the following 3 weeks when patients resumed once-weekly administration. Two or more consecutive missed doses resulted in even lower dulaglutide concentrations, which may be subtherapeutic, and took longer than 3 weeks to return to steady-state after patients resumed their once-weekly administration.
